Cell viability assays are essential laboratory techniques used to determine whether cells are alive, healthy, metabolically active, or undergoing cell death. These assays play an important role in biomedical research, pharmaceutical development, toxicology, cancer research, and drug discovery. By providing measurable information about cellular health, cell viability assays help researchers evaluate how cells respond to drugs, chemicals, biological compounds, environmental conditions, and therapeutic interventions.

What Are Cell Viability Assays?

Cell viability assays are analytical methods designed to measure the proportion of living cells within a population. Depending on the assay principle, researchers may evaluate metabolic activity, membrane integrity, enzyme activity, ATP levels, or other biological indicators associated with viable cells.

Common technologies include colorimetric, fluorometric, luminescent, and dye-exclusion assays. The choice of method depends on the cell type, experimental objective, sample size, and required sensitivity.

Importance in Drug Discovery

One of the major applications of cell viability assays is pharmaceutical research. During drug discovery, researchers need to determine whether a candidate compound produces a desired biological effect without causing excessive cellular toxicity.

Cell viability testing can be incorporated into early-stage screening programs to compare multiple compounds and identify potentially promising candidates. Researchers can also use viability measurements to establish concentration-response relationships and investigate the effects of different treatment durations.

In oncology research, these assays are frequently used to evaluate the effects of anticancer compounds on tumor cells. Researchers may compare treated and untreated cells to determine changes in cellular survival following exposure to a candidate therapy.

Major Types of Cell Viability Assays

Several assay formats are available for laboratory applications. Tetrazolium-based assays, such as MTT and related methods, measure cellular metabolic activity through the conversion of substrates into measurable products.

ATP-based assays use cellular ATP as an indicator of viable cells. Because ATP levels are closely associated with metabolically active cells, luminescent ATP assays can provide sensitive measurements for high-throughput applications.

Dye-exclusion methods, including trypan blue-based approaches, distinguish viable cells from cells with compromised membranes. Fluorescent viability assays can use combinations of dyes to identify living, damaged, or dead cells.

Each method has advantages and limitations. Researchers therefore typically select an assay based on the experimental model, detection platform, throughput requirements, and desired level of sensitivity.
